Organotin(IV) carboxylates and their mixed ligands compounds as potential chemotherapeutics 21 Aug 2018 Organotin Compounds as Anticancer Drugs Organotin(IV) carboxylates having very low IC50 values could be applied for the development of new anti-cancer drugs. However, the low solubility of organotin compounds poses an important difficulty, which may be partially addressed through the use of hydrophilic groups such as flouro and hydroxyl groups. Therefore, we propose to synthesize new organotin carboxylates with more electronegative hydrophilic substituents and their mixed ligand complexes with heterocyclic N-donors such as o-phenanthroline and benzimidazole derivatives in order to individuate a balance between solubility and lipophilicity features in order to achieve efficacy. We propose to screen all compounds by MTT assay. Further, the mechanisms of action of organotins on biological targets are often different or not well known, so new biophysical techniques along with antioxidant enzymes, staining and DNA fragmentation assays need to be applied to understand where the organotin(IV) compounds exert their cytotoxicity. 46.43 Lacs + 3.0 Lacs arrears DBT, New Delhi Prof. P. NMR Structural and Biophysical Studies on binding of anthraquinone derivatives with Guanine-Quadrupl 08 Aug 2018 NMR Structural and Biophysical Studies of anthraquinone derivatives Telomerase enzyme involved in maintenance of telomeres is activated in more than 80% of cancer cells, but is absent in normal cells. So targeting telomerase maintenance mechanism could potentially halt tumor growth across a broad spectrum of tumor types with little cytotoxicity/side effects. Telomeric DNA sequences form four stranded Guanine quadruplex DNA structures. In addition, promoters of oncogenes (responsible for uncontrollable growth in cancer tissues) and other regions in genome contain Guanine quadruplex DNA. Small molecules causing stabilization of their structure by binding could act as effective drugs for anticancer action/gene regulation. We have undertaken a study of interaction of Guanine quadruplex DNA with mitoxantrone and its anthraquinone based derivatives synthesized in our laboratory. We find that binding of mitoxantrone alters circular dichroism, absorption and fluorescence spectra significantly. The external binding of ligands along grooves with binding energy of -6 to -9 kcal/mole is accompanied by hydrogen bond formation leading to thermal stabilization of human and Tetrahymena telomeric DNA by 13 and 6°C, respectively. This may interfere with association of telomerase with telomere and hence telomere dysfunction leading to apoptosis in rapidly proliferating cells. We are presently investigating action of diethyl/dimethyl/amido anthraquinone compounds to improve efficacy of anticancer action. Rs. 44.2272 lakh DBT, New Delhi Prof. Ritu Barthwal: PI
